酶法辅助乙醇优选牡丹种皮总黄酮

Enzyme Assisted Ethanol Extraction of Total Flavonoids from Peony Episperm

摘要: 研究适合于以牡丹种皮为原料提取总黄酮的方法,对其提取方法工艺条件进行优化。考察了酶种类、浓度、酶解孵育温度、孵育时间和pH值等因素对提取工艺的影响,并在单因素实验基础上,利用正交实验法进行工艺优化。考察结果表明在果胶酶0.05 mg·mL-1、酶解孵育温度45℃、pH4.5条件下进行酶解孵育180 min后,总黄酮得率最高可达74.839 mg·g-1,相对于乙醇提取法牡丹种皮总黄酮得率显著提高。

Abstract: We optimized enzyme assisted ethanol extraction of total flavonoids from peony episperm. We studied the effects of factors including enzymolysis temperature, enzyme content, enzymolysis pH, enzymolysis time on the extraction rates of total flavonoids by singlefactor and orthogonal test. The optimal extraction conditions were after enzyming pretreatment under the pectinase content of 0.05 mg·mL-1, enzymolysis temperature of 45℃, enzymolysis pH of 4.5, enzymolysis time of 180 min, total flavonoids yield was up to 74.839 mg·g-1.Compared with the traditional ethanol extraction, the total flavonoids yield was improved significantly.
