INDUCTION, DIFFERENTIATION AND PLANT REGENERATION OF TRIPLOID CALLI OF POPULUS TOMENTOSA

Abstract: Regenerating plants were produced from immature leaves of the triploid of Populus tomentosa by tissue culture on MS basic medium with different plant hormone. The result showed that callus induction frequency reached to 87.6 percent on MS+6-BA 0.5mg/L+NAA 0.1mg/L;differentiation frequency reached to 68.5 percent on 12MS+IAA 0.1mg/L+6-BA 0.1. It can be generated from one to 7.4 axillary shoots on MS+6-BA 0.5mg/L+NAA 0.01mg/L. Rooting frequency of the shoots got to 85.7 percent onMS+IBA 1.0mg/L.
