发根农杆菌介导几丁质酶基因和β-1,3-葡聚糖酶基因转化烟草的研究

THE STUDY ON AGROBACTERIUM RHIZOGENES MEDIATED CHITINASE AND β-1, 3-GLUCANASE TRANSFORMATION FOR TOBACCO

摘要: 用冻融法和三亲交配法将具有几丁质酶基因、β-1,3-葡聚糖酶基因和卡那霉素抗性标记基因的质粒pBLGC导入具有Ri质粒的发根农杆菌中。用叶盘法转化烟草的三个品种,经Kan抗性筛选获得126株再生植株,对119株再生植株分别以启动子CaMV35S、几丁质酶基因和β-1,3-葡聚糖酶基因的引物进行PCR检测,其中几丁质酶基因呈阳性的植株有72株,β-1,3-葡聚糖酶基因呈阳性的有47株,具有几丁质酶基因和β-1,3-葡聚糖酶基因的植株有36株。对转化的24株转基因经PCR-Southern杂交,结果均呈阳性。实验结果表明,外源基因的转化频率与植物品种、外源基因片段的大小有关,几丁质酶基因和β-1,3-葡聚糖酶基因分别或共同整合到植物基因组中。

Abstract: The plasmid pBLGC which constained chitinase gene and β-1, 3-glucanase gene and NTP-II gene (resistansing kanamycin) was transformed into Agrobacterium rhizogenes with root-in-ducing plasmid by using freeze-thaw method and triparental crossing method. Chitinase gene and β-1, 3-glucanase gene were placed under the control of the Cauliflower MosaicVirus 35S promoter and introduced into three varieties of tobacco by Agrobacterium rhizogenes mediated leaf-plated trans-formation method. 126 regenerated plants were selected by Kanamycin. Among them, 119 plants have been tested by 35S, chitinase gene and β-1, 3-glucanase gene primer separatly through PCR elec-trophoresis assay, of which 72 plants showed chitinase gene positive, 47β-1, 3-glucanase gene postive and 36 postive plants containing both chitinase gene and β-1, 3-glucanase gene. The trans-ferring frequency of foreign gene has relation with plant variety and the length of foreign gene. Chitinase gene seemed to inherit easier than β-1, 3-glucanase gene. 24 plants among PCR positive plants have been carried out by PCR-Southern. The result was that they showed positive. The study indicated chitinase gene and β-1, 3-glucanase gene respectively or together introducing and cointe-grating into plant genenome.
