红松成熟胚离体培养器官发生组织学研究

摘要: 红松(Pinus koraiensis sieb.et Zucc)[3]成熟胚离体培养, 先在分化培养基(SH+1-2mg/L BAP+3%蔗糖)培养两周, 然后转入无激素培养基(SH+3%蔗糖), 在下胚轴和上胚轴陆续形成不定芽。石蜡制片、观察红松成熟胚不定芽的器官发生过程。结果表明:不定芽起源于胚轴的皮层细胞, 有的是表皮和皮层细胞同时启动。芽形成全过程可分成四个阶段:启动阶段、芽原基形成阶段、鳞叶原基形成阶段和不定芽结构形成阶段。

Abstract: Cultured 2 weeks in vitro on medium (SH+1-2mg/1 BA+3% sucrose) for differentiation, mature embryos of P.koraiensis were transferred to the medium without hormone (SH+3% sucrose), Adventitious buds pro-duced on epicotyls and hypocotyls continuously, The organogenesis process of adventitious buds was observed with paraffin section method. The results showed that the adventitious:bud arose from cortical cell of hypocotyls, some of them originated from.both cortical cells and epidermis cells.The whole process of adventitious buds development in culture could be.divided into four stages as follows.(1)starting of cells differentiation;(2)bud primordium;(3)shoot apex with needle primordia;(4)adventitious buds.
