濒危植物山白树遗传多样性及亲缘关系的ISSR分析

Genetic Diversity and Relationships of Endangered Sinowilsonia fienryi Hemsl by ISSR

摘要: 利用ISSR分子标记,对我国特有种金缕梅科(Hamamelidaceae)濒危植物山白树(Sinowilsonia fienryi Hemsl)6个野生居群和2个栽培居群75份试验材料的遗传多样性进行研究,以期为该物种种质资源的保护和合理利用提供科学依据。试验结果显示:(1)10条引物共检测到89条清晰的谱带,其中多态性条带85条,多态位点百分率(PPB)为95.5%,说明山白树在物种水平上遗传多样性较高;(2)Nei’s基因多样度指数(H)为0.283 3,Shannon’s多样性信息指数(I)为0.403 4;Nei’s遗传分化系数(Gst)为0.350 7,居群间与居群内变异百分率分别是25.49%,74.51%,说明山白树居群内与居群间都存在遗传分化,且主要遗传分化来自居群内部;(3)聚类分析(UPGMA法)得到,在阈值0.87处,山白树8个调查居群分为三类,分类结果与其地域分布存在一定规律。

Abstract: ISSR-PCR was used to assess the genetic diversity of six wild populations Sinowilsonia fienryi Hemsl, along with two cultivated populations including 75 test materials. We detected 85 polymorphic bands from total 89 bands with 10 primers, and the percentage of polymorphic bands (PPB) was 95.5% indicating that S.fienryi Hemsl had a high level of genetic diversity at species level. Nei’s gene diversity (H), Shannon’s information index (I), the coefficient of gene differentiation (Gst) and the percentage of total variance among and within populations were 0.2833, 0.403 4, 0.350 7, 25.49% and 74.51%, respectively, indicating that there was a significant genetic differentiation both within and between the populations, and genetic diversity was mainly derived from the differentiation within populations. With UPGMA dendrogram, at the specify threshold of 0.87, S.fienryi Hemsl species were separated into three clusters, and most individuals from same habitat were clustered, indicating that there might be a rule between clustering analysis and geographical distribution.
