STUDIES ON PROTEIN CHANGES OF POLLEN MONTHER CELLS DURING MEIOSIS BY MICRO-ISOLATION AND ULTRASENSITIVE SILVER STAIN TECHNIQUES IN LILIUM DAVILL DUCH

Abstract: In this paper, the comparative analysis of protein composition and change in pollen monther cells have been studied during meiosis I cycle by using "Micro-Isolation"、"Micro-Protein SDS-PAGE"and "Ultrasensitive Silver Stain" techniques. The results indicated that more than 25 protein bands could be detected in 100 meiotic cells which were isolated by micromanipulation. Protein composition has not change at prophase I and metaphase I. But 8 bands which have molecular weight 28KD、27KD、26KD、26KD、24KD、23KD、22KD、16KD、, respectively, were disappeared and 185KD protein band was appeared at anaphase I and telophase I. Experiment techniques were set up in this study are very useful for studying protein changes during meiosis and experiment results were obtained in this study are also very valuable for studying biochemical mechanism of meiotic cycle and cycle regulation in higher plants.
