大蒜根尖细胞有丝分裂同步化诱导与中期染色体分离

INDUCTION OF SYNCHRONY AND ISOLATION OF METAPHASE CHROMSOMES FROM MITOTIC ROOT TIP CELLS IN ALLIUM SATIVUM

摘要: 本研究以大蒜(Allium sativum)根尖为材料,用HU-APM双阻断法对细胞有丝分裂中期同步化和前、后、末期部分同步化作了探讨,表明在1.25 mmol/LHU, 4 μmol/APM处理可使细胞有丝分裂中期指数(Met. I)达35%, 18hr的HU单独处理可检测到27%的前期分裂细胞。HU对后、末期的部分同步化也有明显作用,约有17%的后、末期细胞被检测到。用Schubert介绍的方法,分离出中期染色体,每微升悬浮液中含170条染色体。

Abstract: In this study, metaphase cell synchrony and prophase, anaphase-telophase partial synchrony during mitosis of root tip cells in Allium sativum have been studied by using hydroxyurea (HU) and amiprophosmethyl (APM) treatment. It was shown that metaphase index (Met. I) can be increased around 35% at 1.25 mmol/L HU plus 4 μmol/L APM treatments in root tip cells. About 27% prophase cell division can be detected when root tips treated for 18hr with 1.25 mmol/L HU. Partial synchronization of cell division at anaphase-telophase has also been obtained only using HU treatment, mitotic frequency has reached at 17%.
Metaphase chromosomes isolating from root tip cells was performed using Schubert method, about 170 chromosomes were obtained in per-microlitre of the isolation buffer.
