qRT-PCR Analysis of Gene Expression of AG and SOC1 During Flower Development of Fraxinus mandshurica Rupr.

Abstract: We analyzed the temporal expressions of four housekeep genes, 18S rRNA, GAPDH, β-actin and TU, in ash(Fraxinus mandshurica Rupr.) flowers with real-time quantitative PCR. With NormFinder software, TU could be selected as inner reference genes to study the expression of two genes, AG and SOC1, related to the flowering of ash. The expressions of both AG and SOC1 were different between male and female. The expression of AG started with a peak in female during the early period of flowering, followed by a decrease till mature embryo sac stage. In male, the expression of AG reached the peak(1.53) at meiosis stage. The expression of SOC1 in female was lower than that in male. Our results would provide evidence to study the expressions of flowering-related genes during the period of flower development in ash.
