In Vitro Conservation of Rare and Endangered Emmenopterys henryi Oliv. Germplasm

Abstract: The effects of culture temperature, inorganic salts level, sucrose, mannitol and plant growth inhibitor PP333 on in vitro conservation of Emmenopterys henryi Oliv. stems with a bud in MS basic medium were studied using singlefactor experiments and orthogonal experimental method. The results showed that the survival rate of E.henryi Oliv. stems with a bud cultured in 1/2MS medium containing 2.0 mg·L-1 KT, 2.0 mg·L-1 6-BA, 0.1 mg·L-1 NAA, 50 g·L-1 sucrose and 10 g·L-1 mannitol and stored at 9℃ low temperature was up to 80%. PP333 also had a significant effect on in vitro conservation of E.henryi Oliv., the most suitable concentration of PP333 was 6 mg·L-1, the survival rate after 180 d was over 95%. After morphological indexes measuring, physiological and biochemical indexes determination and RAPD analysis detection, the genetic variation of E.henryi Oliv. conservation in vitro plantlets did not appear. The results provided a simple and effective way for in vitro conservation of the rare and endangered plants E.henryi Oliv. germplasm.
