MICRODISSECTION OF CHROMOSOME 1B OF TRITICUM SPELTA VAR. DUHAMELIANCUM AND AMPLIFICATION OF THE CHROMOSOME DNA

Abstract: Objective Gene Rf3 originated from chromosome 1B of Triticum spelta var. Duhameliancum is a fertility-restorer gene for G-type cytoplasmic male sterility. Microdissected chromosome 1B by glass needles from cells in duration of meiotic metaphase. The DNA fragments was amplified by means of LA-PCR. By dot hybridization, the product was confirmed as the amplified DNA from chromosome 1B of T. spelta var. duhameliancum.
