Callus induction and Plant Regeneration of Euonymus microcarpus(Oliv.) Spraggue

Abstract: With the young stems of Euonymus microcarpus(Oliv.) Spraggue as explants, L9(34) orthogonal test was used to study the effects of different sterilization combinations and different basal medium and combinations of different plant growth regulator on the induction and regeneration and rooting of callus. The most appropriate method of surface sterilization on leaves was with 75% alcohol for 30 s, and then 0.1% HgCl2 for 15 min, the optimal medium for callus induction was MS containing 6-BA 3.0 mg·L-1 and 2,4-D 1.0 mg·L-1 with a maximum induction rate of 79%, the optimum medium for shoot differentiation was MS containing 6-BA 2.0 mg·L-1 and NAA 0.2 mg·L-1 with 78.83% of differentiation rate, and the suitable rooting medium was 1/2MS containing NAA 1.2 mg·L-1 with 83.23% of rooting rate.
