Rapid Propagation and Regeneration System of Populus simonii×Populus nigra

Abstract: The young leaves of aseptic seedling from Populus simonii×Populus nigra were used to test the effect of different culture media and plant hormone on adventitious bud induction, rosette buds induction and seedling rooting. The culture media and hormone have the effects on adventitious bud induction and seedling rooting. The best medium of adventitious bud induction from leaves is MS+6-BA 0.5 mg·L-1+NAA 0.05 mg·L-1, the induction rate is 100%, and the best medium of rosette buds induction is MS+6-BA 0.2 mg·L-1+NAA 0.05 mg·L-1. The best medium of seedling rooting has two kinds including MS+NAA 0.25 mg·L-1 and MH+IBA 0.2 mg·L-1 with the rooting rate of more than 99%.
